State sanitary and epidemiological regulation Guidelines for sanitary and epidemiological assessment of the safety and functional potential of probiotic microorganisms used for food production Guidelines MU 2.3.2.2789-10.

These Guidelines establish requirements for microbiological, biochemical, molecular genetic and hygienic evaluation of strains used as starter cultures for the production of probiotic starter cultures, including direct starter cultures application, bacterial concentrates and microbial biomass for the production of probiotic foodstuffs and biologically active food additives, and necessary laboratory research methods for its performance. The guidelines are intended for legal entities and individual entrepreneurs whose activities are related to development, production and circulation of biotechnological products, as well as for testing laboratory centers that carry out control of quality and safety of food raw materials and foodstuffs, and dietary supplements for food, and accredited in the established order for this type of activity. Sensitivity/resistance of strains to antimicrobial drugs determined by the disk diffusion method (DDM) with superimposition standard paper discs impregnated with antimicrobial agents, onto the surface of a solid medium and/or by serial dilution antibiotic in liquid nutrient medium. The method is based on ability of antimicrobial drugs diffusing into the nutrient environment, inhibit the growth of microorganisms sown on the surface agar, around the disks with the formation of zones of growth inhibition, or suppress growth of microbes in a liquid medium with various concentrations added to it drugs. In this case, to characterize the sensitivity strains for antimicrobial drugs set a minimum inhibitory concentration (MIC), at which there is no visible growth of culture. This concentration determines the degree of sensitivity strain to the antibiotic. Establishing a serial dilution method for assessing antibiotic resistance includes the following stages: (a) preparation of antibiotic solutions; (b) preparation of nutrient media with antibiotic solutions; (c) preparation of a suspension of the test strain and control strain, suspension standardization and inoculation; (d) incubation; and (e) interpretation of research results.
